Demonstration of the TAPAS Lable

BioTeZ established a novel surface functionalization kit -the TAPAS system.

The strong interaction between streptavidin and biotin is widely utilized in biotechnological applications (1). The new TAPAS label was developed to extend the portfolio for assay manufactured and will be valuable for its applications across a wide range of biotechnological areas.

To meet the big demand BioTeZ introduced in 2008 the poly-streptavidin a crosslinked streptavidin polymer with increased biotin binding capacity widely used for lateral flow test applications (2). Now is 2018 BioTeZ released a novel surface functionalization platform the TAPAS Labeling Kit. This kit or labeling service allows an effective labeling a target molecules and detection with specific antibody.

The BioTeZ TAPAS Labeling Kit was developed to extend the portfolio for assay manufactured and will be valuable for its applications across a wide range of areas.The BioTeZ TAPAS Labeling Kit is a two component protein-ligand interaction system. One component is the small 1000 Dalton synthetic label called “TA-label”.

The second component is the “PAS-Antibody” which binds the “TA-label” with high specificity and efficiency. The BioTeZ TAPAS Labeling Kit or labelling service allows antibodies, proteins, primer etc. to be covalently labelled with the TA-label. The pre-activated TA-label is designated for the coupling with the target molecules through the reaction with primary amino groups which allows also double labelling strategies ideal for lateral flow test development projects.

“With the BioTeZ TAPAS Labeling Kit we react on the high demand on surface functionalization technologies for the diagnostic field. With our new TAPAS Labeling Kit we support lateral flow test manufacturing and contribute to a more convince to develop a new point of care technology” explains Dr. Janko Brand (Senior Scientist at BioTeZ).

The BioTeZ TAPAS Labeling Kit supports a large number of applications including immobilization, enrichment or detection of antibodies, other proteins and nucleic acids. It can be flexibly combined with various target molecules and detection systems such as fluorescent, luminescent, gold or carbon black (3).

Many commercial immunoassays employ the high affinity interaction between biotin and streptavidin as part of the analyte capture mechanism. As such, these immunoassays are subject to an interference if biotin is part of the diet (4).  The new TAPAS Label is biotin free and will reduce the background and thereby increase the sensitivity for many applications.

“We are glad to demonstrate the ability of the TAPAS Labeling Kit for this important application in the coating and surface functionalization. Currently, we are in dialog with all major distributors and prepare a worldwide roll out of the BioTeZ TAPAS Labeling Kit” says Uwe Ahnert (CEO BioTeZ).

The advantages of the BioTeZ TAPAS Labeling Kit compared to conventional functionalization reagents are the improved signal noise ratio compared to streptavidin and poly-streptavidin. The high affinity binding between TA-Tag and PAS-Capture antibody leads to rapid binding with low cross-reactivity. Since the TA-PAS label is artificial and not present in nature non-specific binding were detected.

Product specifications: The BioTeZ TAPAS Labeling Kit includes the PAS capture antibody, all necessary reagents to label the target molecule with the TA-labels and clean-up columns for purifying the coupling product. For double labelling TA-label and gold nanoparticles it is recommended to do TA-labelling procedure first.

Demonstration of the In Vitro diagnostic ELISA of autoimmune & infectious diseases

Human-diagnostic ELISAs
The ELISA tests listed below consist of 29 kits for In Vitro diagnosis of autoimmune diseases and 2 kits for the diagnosis of a Helicobacter pylori infection.

These ELISA kits are CE marked and come in the standard format of 96 analyses. They all use identical general reagents, namely sample buffer, wash buffer, substrate and stop solution. Hence, one and the same sample dilution may be used for multiple assays; e.g. for simultaneous determination of Cardiolipin IgG and IgM.

All solutions are ready to use (exception: 10-fold wash buffer concentrate) and color-coded. The conjugate color (red, yellow or green) indicates the Ig-class detected, IgG, IgA or IgM, respectively. The color intensity of the blue calibrators reflects their corresponding concentration. The negative control is green, the positive red.

The solid phase features break apart microwells for economic use of the kit. It is packed in a foil bag with zip closure, useful for partial consumption of the solid phase. The frame of the microwell plate displays the parameter name.

The sample is serum or plasma; 10 µL are required throughout. It is diluted 1:100 in the orange coloured sample buffer. 100 µL per microwell is the fixed volume of sample, conjugate, substrate and stop solution.

The tests share one common protocol: 3 x 30 minutes incubation at room temperature and without shaking. Wash steps: unvariably 4 x 350 µL per microwell.

The tests have been validated on the Dynex DS2 system and are accessible to other automated devices. The liquids come in polyethylene bottles with enough volume for automation. The detection system is HRP/TMB, read at 450nm.

The compact outer box is labelled on two adjacent sides for easy test kit identification in fridge stacks. Each product comes with printed Directions for Use (also downloadable), including a Summary Flow Chart on the back, and a kit lot-specific Certificate of Analysis. Most of the tests offer the option of qualitative or quantitative evaluation.

The excellent quality of these products is reflected by a very low number of complaints over the years. Since more than a decade, we regularly participate in several international ring trials (UK: Neqas, US: CAP, Germany: Instand and Referenzinstitut für Bioanalytik) with virtually no deviation of our results from the respective intended response.

Demonstration of the In Vitro diagnostic ELISA of autoimmune & infectious diseases

Human-diagnostic ELISAs
The ELISA tests listed below consist of 29 kits for In Vitro diagnosis of autoimmune diseases and 2 kits for the diagnosis of a Helicobacter pylori infection.

These ELISA kits are CE marked and come in the standard format of 96 analyses. They all use identical general reagents, namely sample buffer, wash buffer, substrate and stop solution. Hence, one and the same sample dilution may be used for multiple assays; e.g. for simultaneous determination of Cardiolipin IgG and IgM.

All solutions are ready to use (exception: 10-fold wash buffer concentrate) and color-coded. The conjugate color (red, yellow or green) indicates the Ig-class detected, IgG, IgA or IgM, respectively. The color intensity of the blue calibrators reflects their corresponding concentration. The negative control is green, the positive red.

The solid phase features break apart microwells for economic use of the kit. It is packed in a foil bag with zip closure, useful for partial consumption of the solid phase. The frame of the microwell plate displays the parameter name.

The sample is serum or plasma; 10 µL are required throughout. It is diluted 1:100 in the orange coloured sample buffer. 100 µL per microwell is the fixed volume of sample, conjugate, substrate and stop solution.

The tests share one common protocol: 3 x 30 minutes incubation at room temperature and without shaking. Wash steps: unvariably 4 x 350 µL per microwell.

The tests have been validated on the Dynex DS2 system and are accessible to other automated devices. The liquids come in polyethylene bottles with enough volume for automation. The detection system is HRP/TMB, read at 450nm.

The compact outer box is labelled on two adjacent sides for easy test kit identification in fridge stacks. Each product comes with printed Directions for Use (also downloadable), including a Summary Flow Chart on the back, and a kit lot-specific Certificate of Analysis. Most of the tests offer the option of qualitative or quantitative evaluation.

The excellent quality of these products is reflected by a very low number of complaints over the years. Since more than a decade, we regularly participate in several international ring trials (UK: Neqas, US: CAP, Germany: Instand and Referenzinstitut für Bioanalytik) with virtually no deviation of our results from the respective intended response.

Putting Ergot Alkaloids where they belong- in the history books

The fungi Claviceps purpurea produces high concentrations of mycotoxins, called ergot alkaloids. Ergot alkaloids are complex molecule with a wide range of harmful biological activities. The new BioTeZ Ergot Alkaloids clean-up columns help to detect and quantify the mycotoxins in crops to prevent the toxification of human, livestock and domestic animals.

Ergot poisoning (ergotism) in humans and domestic animals cause hallucinations, the feeling of itchy, burning skin and can cause effects on neurotransmission and circulation. Ergotism is one of the oldest known human diseases caused by mycotoxins and references to grain diseases have been found in various books of the Bible (850-550 BC). In 1597 scientists at the University of Marburg observed that signs of ergotism often appeared after blighted rye grains were consumed and that ergotism was promoted by cold, damp growing seasons.

In 1630 it was observed that feeding of blighted grain to animals produced an illness similar to human ergotism and by the end of the 18th century poisoning was demonstrated in animals. Finally, in 1764, ergot was recognized as a fungus by Otto von Munchhausen (Schiff, 2006). Until the late 20th century, human and livestock exposure to ergot alkaloids was primarily through ingestion of “ergots,” which are spur-shaped or seed-like resting structures (sclerotia) of ergot fungi, the Claviceps species (Schardl, 2015).

In 1927 more than 11.000 cases of ergotism were reported from Sarapol (Russia). Another incident happened in 1951 when 230 villagers of the popular French tourist town of Pont Saint-Esprit were sickened after ingesting contaminated goods from a local baker. The victims became violently ill with symptoms of severe gastrointestinal upset, dramatic reduction in body temperature, hallucinations and euphoria. Note that ergotamine contains the same dopamine substructure which is also found in psychoactive drugs like lysergic one of the building blocks of the psychoactive drug lysergic acid diethylamide (LSD).

Although reports of ergotism in livestock vary from year-to-year according to rainfall and temperature, most cases occurs in the grain producing areas up today. An increase demand for biological produced products will cause a reduced use of fungicides which will lead to a higher need for ergot alkaloids monitoring in the next years. BioTeZ is already able to deliver a clean-up column of food and feed extracts for the reliable identification and accurate quantification of the predominant ergot alkaloid early in the food chain to avoid ergotism.

The extraction protocol of (Krska et al., 2008) is used in a modified version. Just briefly, a solid-phase extraction and clean-up method based on sodium-neutralized cation exchange to quantify 12 prioritised ergot alkaloids in rye flour and wheat germ oil by HPLC fluorescence analysis. The most abundant ergot alkaloids produced by fungus Claviceps purpurea comprise six pairs of epimeric ergot alkaloids (Köppen et al., 2013).  

In summary ergot alkaloids still play a crucial role in the food and feed industries up to date. The reduction of fungicides in biological natural products will increase the need to monitor the ergot alkaloids in the futures food chain. BioTeZ is starting to develop a new method for directly measuring the primary effect of drugs on their target molecule.

BioTeZ starts the project for the development of a companion diagnostic procedure. The aim is to determine the optimal drug dose for each patient to reduce side effects, to achieve the Therapeutic Window and to increase the effectiveness of the active ingredients by optimizing the dosage (Therapeutic Drug Monitoring).


The project is being funded on behalf of the State of Berlin through the Program for the Promotion of Research, Innovations and Technologies (Pro FIT) and partially funded by the European Regional Development Fund (ERDF). In the next two years, intensive work will be carried out to establish the Drug Primary Performance Monitoring Assay (DPPMA). Drug Primary Performance Monitoring Assay is characterized by the ability to simultaneously determine concentrations across binding curves of two molecules from different classes.

In addition, the application of various detection systems is tested. DPPMA is an excellent addition to BioTeZ’s therapeutic drug monitoring portfolio, with recoveryELISA and others. for Adalimumab and Evolocumab products include for the determination of protein-protein interactions.

BioTeZ established a novel therapeutic drug monitoring assay for Evolocumab and PCSK9

The first PCSK9 Inhibitors reach the market and BioTeZ just completed the development project for the therapeutic drug and antigen monitoring assay. The recoveryELISA® (RPE) allows the simultaneous quantification of the free PCSK9 and the free Evolocumab in one ELISA assay.

Proprotein convertase subtilisin/kexin type 9 (PCSK9) has medical importance since it directly interferes with the lipoprotein particles (LDL) homeostasis. PCSK9 binds to the lipoprotein particles receptor (LDLR) which transports fat molecules within the extracellular fluid. Agents which block PCSK9 can decrease the LDL particle concentrations in the blood and thereby prevent cardiovascular disease.

The first therapeutic monoclonal antibodies against PCSK9 Evolocumab and Alirocumab were approved by the U.S. Food and Drug Administration in 2015 for lowering LDL-particle concentrations when statins and other drugs were not sufficiently effective or poorly tolerated.

For Evolocumab two different dosing regimes are suggested 140 mg every two weeks or 420 mg once per month. By definition Therapeutic Drug Monitoring (TDM) is the clinical practice of measuring specific drugs to maintain a constant concentration in a patient's bloodstream, thereby optimizing individual dosage regimes (1).

“The recoveryELISA® RPE is the only method that simultaneously quantifies the free PCSK9, the PCSK9 neutralization rate and the free available therapeutic antibody Evolocumab in human serum samples during Evolocumab therapy. A pilot study enrolled Evolocumab naïve patients suffering from residual cardiovascular disease, treated for the first time with evolocumab and followed for the next two injections. Serum samples were tested with the recoveryELISA© RPE to quantify free PCSK9 and free evolocumab simultaneously and compared these results with the lipid profile.

“We are glad to demonstrate the ability of the recoveryELISA© PCSK9 & Evolocumab for this important application in a clinical setting. Currently, we evaluate the data and we will start soon with writing up a publication on our findings” says Dr. Janko Brand (senior Scientist at BioTeZ).

The strong interaction between streptavidin and biotin is widely utilized in biotechnological applications (1). The new TAPAS label was developed to extend the portfolio for assay manufactured and will be valuable for its applications across a wide range of biotechnological areas.

To meet the big demand BioTeZ introduced in 2008 the poly-streptavidin a crosslinked streptavidin polymer with increased biotin binding capacity widely used for lateral flow test applications (2). Now is 2018 BioTeZ released a novel surface functionalization platform the TAPASTM Labeling Kit.
