Genekam Zika Virus & Dengue Virus & Chikungunya Virus, Realt

4in1 kit: Genekam has developed a new test (4in1) to differentiate between Zika, Dengue (1-2, 3-4) and Chikungunya in one step. Such assays can be helpful to screen the donors for blood banks as the blood transfusion is one of source of infection, hence it is important to trace the such donors or blood samples, which may be positive to above said viruses. Other applications of such tests are screening of organs, cell cultures and semen donors etc. 4in1 kit can be one of important monitoring tool for mosquito borne pathogens in different clinical and non-clinical settings. Serological tests are of little use as mentioned already. Zika virus is known to cause congenital infections leading to microcephaly, which may cause of number of issues during the whole life of an indivual. Such assays can be used to screen the people in prone areas to know which virus is prevalent as it is same vector Aedes aegypti or A. albopictus, which carry all these viruses. Genekam Test is multiplex assay. Please note that there is technological disadvantage of multiplexing as some time, it is is less sensitive against simplex (if you are running each test as single test). This Genekam assay offer quick screening of samples as it will save money and time. Moreover such tests may be a great help for screening the samples from pregnant women to find whether they are positive or negative. Not only this, this test will indicate the load the viruses also. This test kit contain all components needed to run the assay. User needs to add RNA of the samples into the components and put in the realtime machine and the results can be ready within 2 hours. On one 96 well plate, one can run around 90 samples i.e. one can 90 samples tested within 2 hours.

Samples: blood (even one drop of blood), urine, plasma, serum, nervous tissue, fetal samples and mosquitoes

Contents of the kit: RNA master mix, primers and probes, negative and positive control

Equipments: Realtime PCR machine with minimum 4 different dyes (multiplexing capacity minimum 4)

The important part of this kit is that it differentiates between different Dengue genotypes (1-2 and 3-4) and it can be very important feature as Dengue 2 is known to cause more fatal outcome of the disease.

4in1 kit is being tested with different cross reaction panel e.g. Plasmodium, Westnile virus, JEV, HSV1 and 2, Yellow fever, HCV, HIV, HBV, CMV, Toxoplasma, Influenza, Treponema and so on. They should be negative.

This kit functions excellent as it is highly specific, sensitive and accurate. This kit has been tested with Zika virus reference strain along with strains of Dengue viruses and Chikungunya viruses. This kit detects all these strains. Virtual analysis of primers and probes show that they are highly specific for their targets and does not cross reaction with any pathogens or part of human body.

Isolation of circulating tumour cell kit with two markers and PCR Kit for Biomarkers of Melanoma from CTC


Human circulating tumour cells (CTC) in blood are an important indicator of the progression of a cancer and in some cases, these may be an indication of so called sleeping tumour cells i.e. it does mean that there is an active disease. Therefore PCR tests are being conducted in many laboratories around the world for presence of markers of such blood cells. As the number of such cell is very low, therefore there is a need of enrichment of these cells before doing RNA isolation. Genekam Biotechnology AG has developed this enrichment step and this can be combined with mini-column method or other method. Genekam offers this enrichment step as sterilised product because some users may want to grow enriched cells in order to do other studies including nucleic cells isolation e.g. tumour resistance (if possible!), but that one can go directly to RNA isolation step after enrichment even under unsterilised environment in order to run the tumour marker PCRassay. There is only 1 circulating tumour cell in one million of mononuclear cells, therefore enrichment is very important step during the conducting PCR for human cancer markers from blood. Peripheral blood is one of easiest way to access against the biopsy of lymphnodes or bone marrow, hence it is an excellent idea, if one can develop more and more assays based on CTC.

Important feature of enrichment step is that one can do the isolation of circulating tumour cells directly from blood 1ml or 3ml and there is no need of using density gradient method.

Genekam CTC Enrichment kit is very simple to use: There are only 3 steps:

isolation of all cells from 1 or 2 ml of blood to develop a pellet
adding of the beads to this pellet

Dengue Fever Virus (Realtime PCR-Kit) FR074

Dengue virus is a member of family Flaviridae and is single stranded enveloped RNA virus. It has 4 subtypes named as Dengue-1, Dengue-2, engue-3 and Dengue 4. Dengue virus infection is transmitted through bites of vector, which is a mosquito named Aedes aegypti. This infection is common in rainy season in tropical countries and subtropical regions of the world. The dengue virus is becoming a major threat during the last decade and it is rising at very alarming rate. The spread of Dengue virus is accelerated through changes in climate, which is caused through too much production of carbon dioxide. Dengue virus is infecting around 400 million people around the world. According to WHO reports, there were confirmed 10 million dengue virus infection in 2010 around the world. Recently (2014) the dengue virus outbreak is reported in Japan. Incubation period is 4-10 days after the bites of mosquitoes. Dengue infection results in fever along with symptoms like chilling, headache, pain behind eyes, joint pains, vomiting etc. The symptoms last for 2-7 days. It effects children, adults and with self limiting disease, but in some case, the disease is fatal as severe form of dengue infection is caused through plasma leaking, fluid accumulation, bleeding etc. Warning signs occur after 3-7days of start of infection symptoms and these are abdominal pain, vomiting, blood in vomit. At this stage, there is a need of medical care to avoid the fatality. There is loss of thrombocytes, hence platelets transfusion is given. Dengue virus can be diagnosed with serological and molecular biological methods like PCR. PCR method has advantage over their serological methods, which are prone to cross reactions leading to false results. PCR method can be used to differentiate the different strains of dengue viruses. PCR method should be used to differentiate between Dengue virus types and other mosquito borne diseases like malaria, yellow fever virus etc. There is no vaccine and no treatment available for this virus, hence prevention and supportive treatment like fluid therapy, platelet transfusion etc as maintaining the body fluid can reduce the risk of mortality. PCR technology should be used to monitor the presence of virus in mosquito. In case of positive population, mosquito eradication should be used to reduce the infected mosquito population.

NoMore Mycoplasma solution (Sterile) (sufficient to make 200 ml cell culture media) (STEM23)

This solution is 100X, hence it can be diluted with your media 1: 100 (1 Part of No More Mycoplasma solution + 99 Part sof your cell culture media e.g. user needs 10 ml of Media (100 ul of Genekam No More Media to 9.9 ml of cell culture Media). The concentration can be increased depending upon the degree of Mycoplasma infection e.g. 50X i.e. 200 ul of No More Mycoplasma solution in 9.8 ml cell culture media to generate 10 ml of cell culture media. The working solution of cell culture may be stored for 2-4 weeks, but it is better to make it fresh.

Mycoplasma are common problem of cell cultures. There are different types of mycoplasmas, which may infect cell cultures e.g. in case you are generating a cell line from mouse, Mycoplasma may be of mouse origin. It may come from human beings, which are interacting with the cell cultures e.g. Mycoplasma pneumoniae. They may come from serum being used e.g. fetal calf serum e.g. Mycoplasma bovis. The common infecting agents in cell cultures are M. arginini, M. frementans, M. hominis, M. hyorhinis, M. orale and Acholeplasma laidlawii. Penicillin group does not remove mycoplasma infection. The biggest problem with mycoplasma contamination is that they are aerosol transmission i.e. they are in air. In order to treat the infected cell lines, these guidelines should be followed:

1. Clean the working area with disinfectant and UV light.

2. To treat the infected cell cultures: a part of infected cell cultures should be freezed and other part of this should be treated as during the treatment, it may be possible that one can lose the cell cultures. If user has frozen the cell culture, it can be thawed in case that treatement was not successful.

The treatment should be done for the period of 14 days or the duration of treatment can be reduced depending on the severity of infection.During the treatment, one should observe the cell cultures. Please take precautions during working as one is working with live pathogens.

NoMore Mycoplasma solution contains substances, which are very effective active against a large number of mycoplasma strains.

Prepare the media while adding 10 ml of Genekam osteogenic differentiationmedia into 90ml of DMEM containing 10 % FCS + 1% SP + 1% Glutamine or in your culturing media. Store it at 4°C. You can prepare less media, but the ratio should be 1:10.


After you have stem cells e.g. MSC. To isolate MSC cells, one can use Genekam Stem cell isolator kit. After you get 60-85% confluence during culturing of stem cells, please remove the cell using trypsin or Genekam cell remover. and wash the cells to make free from trypsin while Centifuging 1400 rmp for 5 minutes and suspend the pellet now to make the cell count using trypan blue.
Now add the cells 3-5 x 103 / cm2 on a plate e.g. 9 or 12 wells in osteogenic differentiation media (containing DMEM + 10 % FCS or other serum source + Genekam osteogenicc differentiation media) for two weeks while feeding the cells 3-5 days with this media for 3 weeks.
After two weeks, you can stain with cells with Alizarin Red S to see the bone formation.

After you have stem cells e.g. MSC. To isolate MSC cells, one can use Genekam Stem cell isolator kit. After you get 60-85% confluence during culturing of stem cells, please remove the cell using trypsin or Genekam cell remover and wash the cells to make free from trypsin while Centifuge at 1400 rpm for 5 minutes and suspend the pellet now to make the cell count using trypan blue.
Now add the cells 0.5 x 105 / ml in chrondrogenic differentiation media (containing DMEM + 10 % FCS or other serum source) in 15ml sterile tube and centrifuge it at 1200 rmp for 5 minutes. The pellet will be formed. Now keep the tube in CO2 inocubator at 5% CO2 for two weeks (donot distrub the pellet; donot tight (keep it loose!) the cap of tube in order to low the exchange of CO2) while feeding the cells 3-5 days with this media for 2 weeks.
After two weeks, you can stain with cells with Toludine blue to detect the secretion of sulfated glycosaminoglycans or stain with safranin O staining.

This media is tolerated well with blood cells e.g. B and T-lymphocytes, monocytes, stem cells etc. One can use it to store cell lines like hybridoma, cancer lines and routine cells. If you are using your own formulation or formulation from other companies, please use always two different media to store the cells in order to have better recovery rate. Our attempts will be to offer the innovative products on the market to accelerate the stem cell research in order to improve the quality and safety standards of stem cell solutions. STEM01 is ready to use formulation, where user need to add the cells in the freezing solution and freeze them at -20 degrees for a weeks or in liquid nitrogen or dry ice (slow freezing!). Genekam Biotechnology AG has developed a new method to isolate the mononuclear cells (MNC) from different sources like bone marrow aspirates, buffy coats, blood, umbilical cord blood and other sources. Usually one uses density gradient centrifugation, which is a time consuming and need a lot of precautions during the pipetting of fluids on density gradient. Making a mistake means that you are going to lose the whole samples. Density gradient methods are not good for isolation of MNC from the sources, where you have less than one ml volume and more you get only a part of MNC as you cannot take out the cells from white ring. Genekam Biotechnology AG has developed one method, where you can isolate all cells during your isolation available in sample within 20 minutes. Not only this method is faster, but it is also economical and let you isolate the MNC from as little as 25µl of samples along with the larger size samples. This is a sterile product.

Genekam Quick MNC isolator is ready to use sterile solution for isolating the mononuclear cells (MNC) quickly from blood, buffy coats, bone marrow aspirates, spleen etc of different mammals like human, rat, primates, cattle, sheep, goat, pig, horse, camel, dog, cat etc.

Here are a few examples of application of Quick MNC isolater:

A. Isolation of MNC from human peripheral blood or buffy coat:


Add 900µl of Genekam Quick MNC isolater to each tube containing up to 100 µl of whole blood or buffy coat in one sterile tube as this is ration (1:5) of solution to the sample e.g. if you want to isolate MNC from 2ml of buffy coat and add 8 ml of Quick MNC isolator. You have to decide howmuch blood or buffy coat sample do you want to use to isolate the MNC as you will get a lot of MNC against the conventional method of density gradient centrifugation.
Warning: Kindly work with smallest volume of sample to gain some experience with this kit instead of destroying whole sample for first time.
Gently vortex each tube immediately after adding the solution. Keep at room temperature, protected from light, for 7-10 minutes.
Centrifuge 1600 rpm for 5 minutes. Aspirate or discard supernatant without disturbing pellet.
Resuspend the pellet in the appropriate buffer (usually it is PBS in most of applications) and wash 2X while centrifuging for 1600 rmp with brake for 5 minutes each.
Resuspend the pellet and proceed with further procedures e.g. counting the number of MNC and go for your analysis e.g. cell culturing.
B. Isolation of MNC of human bone marrow aspirate:


Add 400µl of Genekam Quick MNC isolater to each tube containing up to 200µl of human bone marrow in one sterile tube as this is ration of solution to the sample (1:3 ration) e.g. if you want to isolate MNC from 2 ml of bone marrow aspirate and add 4 ml of Quick MNC isolator. You have to decide about the volume of bone marrow you want to use to isolate the MNC because you will get a lot of MNC against the conventional method of density gradient centrifugation.
Warning: If you do not have any experience with this kit, kindly use smallest volume of sample to have experience with this kit before you destroyed the whole sample as you may have to adjust ration e.g. you have still erythrocytes at the end, you have to use more MNC isolator (1 volume sample and 4 volume MNC isolator e.g. 200 µl sample and 600 µl of MNC isolator) or wash one time more. Usually the above said protocol is good enough to isolate MNC.
Gently vortex each tube immediately after adding the MNC isolator solution. Keep at room temperature, protected from light, for 7-10 minutes.
Centrifuge 1600 rpm for 5 minutes. Aspirate or discard supernatant without disturbing pellet.
Resuspend the pellet in the appropriate buffer (usually it is PBS in most of applications) and wash 2X while centrifuging for 1600 rmp with brake for 5 minutes each.
Resuspend the pellet and proceed with further procedures e.g. counting the number of MNC and go for your analysis e.g. cell culturing.

C. Isolation of MSC from of mouse spleen:


Prepare a single cell suspension after harvesting the mouse spleen.
Pellet the cells by centrifugation (1600 rpm); discard the supernatant.
Now suspend the pellet in 5 ml of MNC isolator isolation.
Incubate at room temperature for 7 minutes with occasional shaking.
Add to MSC isolator solution with spleen cell with 20-30 ml of PBS buffer.
Spin the cells (1600 rpm) for 5-7 minutes and discard the supernatant.
Resuspend the pellet in the appropriate buffer (usually it is PBS in most of applications) and wash 2X while centrifuging for 1600 rmp with brake for 5 minutes each.
Resuspend the pellet and proceed with further procedures e.g. counting the number of MNC and go for your analysis e.g. cell culturing.

Genekam Stem cells isolater from Umbilical blood samples: Genekam Biotechnology AG has developed a simple, economical method to isolate stem cells from umblical cord. You just add the solution into umblical blood and centrifuge it. You can isolate the stem cells from small volumes of fluid also e.g. 100 µl (provided they contain the stem cells) as well as larger size samples. Now you can isolate the stem cells with different methods as well as you can do further analysis. This method is far better than density gradient method to isolate the stem cells.
Genekam Stem cell (SC) isolator from umbilical cord blood is ready to use sterile solution for isolating stem cells containing populations (SCP) quickly from blood, buffy coats, bone marrow aspirates etc of different mammals like human, rat, primates, cattle, sheep, goat, pig, horse, camel, dog, cat etc.

Here are a few examples of application of SC isolater:

A. Isolation of SCP from umbilical cord blood:

You have to decide howmuch blood sample do you want to use to isolate SCP as you will get a lot of SCP against the conventional method of density gradient centrifugation.
Warning: Kindly work with smallest volume of sample to gain some experience with this kit instead of destroying whole sample for first time.
Gently vortex each tube immediately after adding the solution. Keep at room temperature, protected from light, for 7-10 minutes.
Centrifuge 1600 rpm for 5 minutes. Aspirate or discard supernatant without disturbing pellet.
Resuspend the pellet in washing buffer and wash 2X while centrifuging for 1600 rmp with brake for 5 minutes each.
Resuspend the pellet and proceed with further procedures e.g. counting the cell number and go for your isolation of stem cells e.g. cell culturing or magnetic beads.
Stem cells can be isolated through pouring the isolated SCP (1x 106/cm2 suspended in DMEM with 10-15% FCS or other media; it is recommended to use FCS precoated plates as they may increase the chances to get the stem cells) into cell culture plates. Keep them at 37°C and 5% for 24-72 hours. After that discard the supernant as the stem cells will be adherent on plastic as adherent fibroblastiod cells, they can be grown and subcultured. Subconfluence of these cells can be isolated with trypsin. Usually cells at the second passage and thereafter can be replated 3.5 to 5 x 103/cm2.

B. Isolation of SCP from human bone marrow aspirate:

Add 400 µl of Genekam SC isolater to each tube containing up to 200 µl of human bone marrow in one sterile tube as this is ration of solution to the sample (1:3 ration) e.g. if you want to isolate SCP from 4 ml of bone marrow aspirate and add 8 ml of SCP isolator. You have to decide about the volume of bone marrow aspirate you want to use to isolate the cells because you will get a lot of cells against the conventional method of density gradient centrifugation.
Warning: If you donot have any experience with this kit, kindly use smallest volume of sample to have experience with this kit before you destroyed the whole sample. You may have to adjust ration e.g. you have still SCP containing some erythrocytes at the end, you have to use more SC isolator (1 volume sample and 4 volume SC isolator e.g. 200 µl sample and 600 µl of SC isolator) or wash one time more. Usually the above said protocoal in point 1 is good enough to isoate.<

Human Tumour marker for Breast cancer (Gel agarose based PCR)

Usually biospies are standard method in breast cancer for detection of tumour. Biopsies are painful methods. But there is no way or method to find whether the cancer cells are underway in the whole body, therefore blood samples offer an unique oppurtunity to detect the tumour cells. These methods are very important to detect them to find whether cancer cells are in blood before or after the therapy. Genekam Hypo-osmotic swelling test (HOS Test)

Genekam biotechnology AG has developed a test called hypo-osmotic swelling test (HOS). which can be used for evaluation of sperm membrane intergrity as a functional membrane is important to know status of fertlity.

The HOS test determines ability of sperm member to maintain an equillibrium between the sperm and its environment. During HOS test, there is influx of fluid in sperm, which leads the sperm tail to be coiled and balloon (swelling).
