New findings on short-term cultivated CAR T cells in cancer treatment

Traditional CAR T cell production is a lengthy process that can delay treatment. To address this, a new method involving short-term cultivation of CAR T cells has been developed, allowing for quicker patient treatment. However, this method raises safety concerns, particularly the risk of cytokine release syndrome (CRS), a serious complication where excessive immune proteins cause systemic inflammation, potentially leading to organ failure.

A team led by Professor Christian Buchholz at the Paul-Ehrlich-Institut conducted studies using mouse models and cell-based tests to evaluate these risks. Their research indicates that short-term cultivated CAR T cells have a higher propensity to trigger CRS than conventionally cultivated cells. This risk is attributed to residual lentiviral vector particles used in the genetic modification process.

The findings highlight the need for careful risk assessment and management strategies before clinical application. Establishing direct test systems to evaluate the safety of these rapidly produced CAR T cells is crucial. This will ensure that the benefits of quicker availability do not outweigh the potential risks.
